Components of an HPLC system

Every HPLC system contains at least five elements:

• Pump (several pumps are also possible)
• HPLC column
• Injector for the flow medium
• Detector
• Computer for controlling the system

The running medium is fed as a mobile phase through a valve into the piston chamber of the pump and finally transported further through the pump. After the sample has passed through the separation column, the detector detects it as it exits and passes the data to the computer for processing.

The size of the pores and particles also plays an important role in the selection. At WICOM you will find HPLC columns with particle sizes up to 60 µm, while for the pores there are mostly standard sizes of 3µm, 3.5µm, 5µm and 10µm. Avoid damage

In order to avoid premature replacement of elements of the apparatus, there are a few precautionary measures that you can take care of yourself.
Do not allow pressure to exceed 400 bar, as this can cause damage/leakage. Your equipment can become clogged if you leave buffers in it. If you tighten fittings too much, they may break off or lose their tightness. In addition, you should never store RP columns in water as this can cause bacteria to multiply.
In general, it is important to work calmly and with concentration while carrying out the HPLC technique and never fall into uncontrolled actionism. This also applies if an execution does not go as planned. By using them thoughtfully, the individual components can be used for a long time before a spare part becomes necessary.
