Columns are now available for a wide variety of chromatography modes. Therefore, it is important to consider both the method and its phases when selecting a suitable HPLC column. For example, there are HPLC columns for normal phase, size exclusion, affinity, reversed phase and ion exchange chromatography. The product types are designed to meet the corresponding requirements of the modes and optimally support you in carrying out your method, because depending on the phase, the columns have different properties (depending on the phase, for example, non-ionic, water-soluble, etc.).

Normal phase and reversed phase chromatography

A distinction is made between normal phase and reversed phase chromatography techniques.
The normal phase (NP) technique was the most common for a long time, which is why the corresponding HPLC columns can also be found in the store from many different manufacturers. Crucial for this methodology is the stationary phase with polar characteristics of the surface as well as the mobile phase with non-polar characteristics. Since many different solvents can be used here, the NP technique is very powerful, but has a disadvantage due to its complexity.
The ratios of polarity are reversed in the reversed-phases (RP) technique as the name suggests. Thus, the stationary phase is now non-polar and the mobile phase is polar. In this process, the nonpolar chains of the RP columns are bonded to a silica gel scaffold or polymer, resulting in hydrophobic properties. Since this process is now the most popular, a range of columns exists from all manufacturers.

Material

Benefit from HPLC columns made of different materials in our store. When choosing the material, you should pay particular attention to the properties of the analyses. Conventional columns of this type usually have the following processing:

  • Diol

  • Phenyl

The materials currently used are already much more robust and durable than those used in the early days of HPLC in the 1960s. The quality of the stationary phases has been significantly improved.

Pore and particle size

The HPLC columns in our online store have different pore and particle sizes. There are columns with particle sizes from 1.5µm to 60µm. In general, our products have particle sizes of 3µm, 3.5µm, 5µm and 10µm.

SFC columns

A cost-saving and flexible alternative: the SFC method (SFC = Supercritical Fluid Chromatography or supercritical fluid chromatography) allows environmentally friendly CO2-based separation of substances. Preparative columns with an internal diameter of 10 to 50 mm and different particle sizes are used here, resulting in particularly rapid separation of substances. In addition, the process is particularly inexpensive due to the reduced solvent consumption. Thus, the SFC method offers several advantages over the HPLC technique. Deviations according to the USP

The USP (Unites States Pharmacopoeia) is one of the most important pharmacopoeias worldwide. And defines fixed standards for the quality of food and drugs. The following deviations are allowed according to the USP:

  • Inner diameter of the column: variable, if flow rate is constant
  • Length of column: +/- 70%
  • Flow rate: +/- 50%
  • Size of particles: -50%
  • Component ratio (mobile phase): +/- 30%
  • ph-value (mobile phase): +/- 0.2
  • Temperature of the column: +/- 10 degrees Celsius
  • Salt concentration (in buffer): +/- 10%
  • Wavelength (UV-Vis detector): +/- 3 nm
  • Injection volume: reduction until detection limits are reached
